Primary human monocytes were separated from peripheral blood by counter-flow elutriation. They were harvested, lysed and their protein profiles analyzed by two-dimensional (2D) gel electrophoresis. The overall pattern of proteins obtained with this method was different from that of monocytes of other cell sources. Therefore, the physiological meaning of the proteome of primary monocytes is still unclear. Nevertheless, the data showed that procoagulant activity is up-regulated on activation, and that the mechanisms of its regulation differ from that of endothelial cells and monocytes. Moreover, the function of monocyte lipopolysaccharide-induced proinflammatory cytokines was analyzed using 2D gels. The results indicated that the pattern of proteins expressed upon activation of monocytes was different from that of endothelial cells, and that the molecules detected by these cells may be different. The 2D gel method, which can be used to analyze the subtlety of cell functions, may be useful for analyzing and comparing physiological functions of cells.Intel's 3D XPoint Memory is Here, and a Lot of Rumors Are Flying
